EFJ5100 VHF Handheld

I have a vhf 5100 hand held with 256 channels, is there any way to enable the option for it to use 512 channels. Every time I try to enable it through the software it tries to find a file on my PC.

You would have to purchase the option from EFJ, the file the program is looking for is an options file to turn on/off features that can only be generated at the factory.
Current list price to upgrade for 256 to 512 TGs is around $200.

would that include upgrading the radio to the latest firmware?

No, but the code updaters only run about 20 bucks and you can do as many radios as you want with it.
